How do I choose between before, after, and while with -ing?

Use "after + -ing" when the -ing action is completed first: "After finishing the report, he sent it." Use "while + -ing" when two actions happen at the same time: "While driving home, he listened to a podcast." In exercises, time clues usually reveal the order. If the sentence describes simultaneous actions, "while" is normally the best choice for meaning in that sentence.

What is the difference between after doing and on doing?

"After doing" simply shows sequence: one action is completed, then another action follows. "On doing" is more specific; it means immediately when something happens or as soon as someone does something. "After reading the email, she called her colleague" tells us the reading came first. "On reading the email, she called her colleague" suggests the call was an immediate reaction. Both can sometimes be correct, but "on" adds a stronger sense of instant response, which is why it often appears with news, alarms, arrivals, and discoveries.

How do instead of and in spite of work before participles?

Both are followed by an -ing form, but they express different ideas. "Instead of + -ing" shows an alternative action: "Instead of taking the bus, she walked." One choice replaces another. "In spite of + -ing" shows contrast: "In spite of being tired, she continued working." The result is surprising because the first idea would normally point the other way. A useful test is to ask whether the sentence means "rather than" or "although". "Rather than" points to "instead of"; "although" points to "in spite of" in context.
